Numerous approaches have been taken exploiting differences in cell density, membrane properties, phagocytic abilities, and autofluorescence. Of these the most widely used is the density difference between eosinophils and other cells. Presently, the mostly widely used method is sedimentation through Metrizamide although sedimentation in polyvinylpyrrolidone-coated silica particles (Percoll) also provides highly purified eosinophils in good yield.
